Association of Apolipoprotein E ε4 with Tau and Amyloid‐β PET in a memory clinic cohort

چکیده
Background The Apolipoprotein E ε4 (ApoE4) is the strongest genetic risk factor for Alzheimer’s disease (AD). Previous studies have found a link between ApoE4 genotype and tau (T) deposition in medial temporal lobe (MTL). However, further disentangling influence of on amyloid (A) are still needed. aim this study was to assess depending subject memory clinic cohort. Method 89 subjects from Geneva Memory Center underwent PET imaging, ApoE genotyping (ApoE2 carriers, ApoE3 homozygotes, carriers). remaining were classified as ApoE3. Amyloid uptake has been calculated centiloid values standardized value ratio (SUVR), normalized cerebellar uptake. Positivity each biomarker based thresholds published by previous studies. Correlation SUVR general calculated. ANOVA with Tuckey correction multiple comparisons used differences or across different genotypes. Result 85% A+T+ carriers gene. association neocortical MTL stratified showed that these measures correlated (r = 0.52, p 0.001). correlation not significant non-carriers. higher significantly when compared Furthermore, lateral superior lobes also vs. non-carriers even considering value, showing an amyloid-independent limited MTL. Centiloid similar comparing Conclusion gene present increase both load respect increased independent MTL, entire pathology carriers.

Frequency of the Apolipoprotein E ε4 Allele in a Memory Clinic Cohort in Beijing: A Naturalistic Descriptive Study
OBJECTIVES To describe the distribution of apolipoprotein E (APOE) genotypes among an elderly Chinese patient population with memory complaints treated in a memory clinic in Beijing and to compare the ε4 allele frequency among individuals with subjective cognitive impairment (SCI), mild cognitive impairment (MCI) and Alzheimer's disease (AD). METHODS A total of 385 subjects with memory compla...
متن کاملDepression was associated with apolipoprotein E ε4 allele polymorphism: A meta-analysis
Objective(s): Previous studies have reached different conclusions regarding an association between apolipoprotein E (APOE) polymorphisms and depression. This meta-analysis was designed to clarify these controversies. Materials and Methods: Literatures were identified reviewing the national and international databases. The eligible articles for meta-analysis were determined by quality assessment...
متن کاملA FDG-PET Study of Metabolic Networks in Apolipoprotein E ε4 Allele Carriers.
Recently, some studies have applied the graph theory in brain network analysis in Alzheimer's disease (AD) and Mild Cognitive Impairment (MCI). However, relatively little research has specifically explored the properties of the metabolic network in apolipoprotein E (APOE) ε4 allele carriers. In our study, all the subjects, including ADs, MCIs and NCs (normal controls) were divided into 165 APOE...
متن کاملAge-Related Association between Apolipoprotein E ε4 and Cognitive Function in Japanese Patients with Alzheimer's Disease
AIMS In the present study, we investigated whether apolipoprotein E (APOE) polymorphisms influenced the cognitive function of Japanese patients with Alzheimer's disease (AD) at certain ages. METHODS Among 200 outpatients with dementia and amnestic mild cognitive impairment, 133 Japanese patients with AD were recruited and divided into two genotypic groups: APOE ε4 carriers and noncarriers. Th...
